    A currently "selected" spot will display as Orange.

    You can use Delete for that so long as you are working with the spot tool as the current "focus" window (to use a Windows term).

    If you open the spot tool as a floating tool on your screen you will see the rest of the controls including a "-" box. If you click on that the currently selected spot will be deleted.

    You do indeed seem to be completely correct that the minus key does nothing.

    Remove spot is not pre-defined in keyboard short cuts and defining it seems not to work either in V10.1 .

    It is probably worth raising a support case.

    ETA. You can also move a spot and change its size. If you are not getting the expected results but believe the spot shout be responding to treatment try those options as well.

    Thanks SFA. I didn't realize the spot tool could be opened as a floater - that solves the problem.
