    You can do that, but not with the black and white tool, which is one of the few that doesn't work on a layer. You can create a layer and desaturate it but erase the mask from areas you want to be in colour.

    Thanks for the reply. Although you misunderstood my question, your reply did tell me that layer doesn’t work in Black and White.

    You do have some potential for something like split toning techniques on the B&W tool AND through mask enabled Color tools. That might not be exactly what you are seeking.

    If Spot Color is your thing that is also possible  - some included Styles offer such a facility and might provide some insight about possible approaches to that option.
